Themonoclonalantibody31reactsspecificallywithsecretoryleukocyteproteinaseinhibitor(SLPI;alsoknownasantileukoprotease(ALP)).SLPIisa11.7kDacationicinhibitorofneutrophilelastaseandtoalesserextentofcathepsinG.Itislocallyproducedbyepithelialcellsinthelung,skinandotherorgansandbyPolymorphonuclearleukocytes(PMN)and(inmice)bymacrophages.Inadditiontoitsproteinaseinhibitorypropertiesthatmayservetoprotectagainstproteolyticinjury,itwasrecentlyshownthatSLPIalsodisplaysseveralotherfunctionssuchasantimicrobialandantiinflammatoryactivities.TheseappeartobeindependentofitsABIlitytoinhibitPMNserineproteinases.SLPIhasalsobeendemonstratedtodisplayantibacterialandantifungalactivityatconcentrationsinwhichSLPIispresentinmucosalsecretionsincludingthoseofthelung.AnotherpossIBLeroleforSLPIisinhibitionofproteindisulphideisomerasethatisconsideredessentialforinvasionofacellbytheHumanImmunodeficiencyVirus(HIV).Thereactivityoftheantibody31withisolateddomainsofSLPIwasevaluatedusingdomainsobtainedbycleavageusingpartialacidichydrolysis.Duetothisfactthemonoclonalantibody31recognizesotherSLPIcleavageproducts.
