ThemonoclonalantibodyH398recognizestheextracellularpartoftheTumorNecrosisFactorReceptortypeI(TNF-RI)ofthemembrane-boundaswellasthesolublereceptor.TNF-RI(~55-60kDa)ispresentonmostcelltypesandisconsideredtoplayaprominentroleincellstimulationbyTNF-alpha.TNF-alphaactivatesinflammatoryresponses,inducesapoptosis,regulatescellularproliferation,andmayevenpromotecancerprogression.TheeffectsofTNF-alphaaremediatedbyTNF-RIandTNF-RII,whichhavebothdistinctandoverlappingdownstreamsignalingcascades.InductionofcytotoxicityandotherfunctionsaremediatedlargelyviaTNF-RI.TNF-RIisequallywellactivatedbyboththe17kDasolubleand26kDamembrane-boundform,whereasTNF-RIIisefficientlyactivatedonlybythemembraneboundformofTNF-alpha.TNF-RIsignalingisinitiatedwhentrimericTNF-alphabindsTNF-RIreceptors.SubsequentTNF-RItrimerizationpromotestherecruitmentofaproximalsignalingcomplexcomposedofTNFReceptorAssociatedproteinwithaDeathDomain(TRADD),ReceptorInteractingProtein(RIP),cellularInhibitorofApoptosisProtein1(cIAP1),TNFReceptorAssociatedFactor2(TRAF2),andlikelyTRAF5.StudieswithTNF-RI-deficientmiceindicatethatTNF-RImediatesmostoftheproliferation,pro-inflammatory,andapoptosis-activatingpathways.
