MannoseBindingLectin(MBL)alsocalledmannose-ormannan-bindingprotein(MBP)isamemberofthecollectinsandanimportantelementininnateimmunity.MBLisanoligomericlectinthatrecognizescarbohydratesasmannoseandN-acetylglucosamineonpathogens.MBLcontainsacysteinerich,acollagenlikeandacarbohydraterecognitiondomain.ItformsacomplexwithC1r/C1slikeserineproteasesdesignatedMASPthatproteolyticallycleaveC4,C2andC3.MBLisabletoactivatethecomplementpathwayindependentoftheclassicalandalternativecomplementactivationpathways.TheMBL-MASPpathway(betterknownasthelectinpathway)isantibodyandC1q-independent.MBLexhibitscomplement-dependentanti-bacterialactivityandactsdirectlyasanopsonicandthereforeplaysanimportantroleininnateimmunity.MBLissynthesizedbyhepatocytesandhasbeenisolatedfromtheliverandserumofseveralvertebratespecies.OnlyoneformofhumanMBLhasbeencharacterized,whiletwoformsarefoundinrhesusmonkeys,rabbits,ratsandmice.ThemurineformsareknownasMBL-AandMBL-C.TheMBL-Cconcentrationsinserumareabout6-foldhighercomparedtothatofMBL-A.MBL-A,butnotMBL-CwasfoundtobeanacutephaseproteinincaseinandLPS-injectionmodels.MBL-CexistsinhigheroligomericformsthanMBL-A.InplasmaofhealthymiceMBL-Cconcentrationsrangefrom16to118µg/ml.IninfectiousdiseasestheMBL-Cconcentrationwasnotfoundtoincreasesignificantly.
