Antimicrobialproteins(AMP)areafundamentalelementoftheprimaryresponseagainstpathogens.AMP’saresmallendogenouscationicmoleculesexpressedbyphagocyticandepithelialcells.TheantimicrobialactivityofAMP’sisdirectedtowardsabroadspectrumofpathogens,likeGram-positive&negativebacteria,viruses,yeastandfungi.AMP’saidininnateandadaptiveimmunityviadirectinactivationandbyimmunomodulatoryactivitylikeleukocytemigration.DefensinsarethemostprominentmammalianAMP’s.Threedefensinpeptidefamiliesareidentified,theα-,β-,andθ-defensins.Theyarecharacterizedbyatriple-strandedβ-hairpinstructure,sixdisulfide-linkedcysteineresiduesandapositivecharge.Theyaresynthesizedaspreproteinsandundergoprocessingtobecomeafullyactivepeptide.Defensinsaredividedinalpha-andbeta-defensinsdependingontheirdisulfidebridgingpattern.α-andβ-defensinsmodifycellmigrationandmaturation,inducecytokinesandtriggerhistaminandprostaglandinD2releasefrommastcells.Therearesixα-defensins:HNP(humanneutrophilpeptide)-1to-4,HD5&6.Humanbeta-defensin1(hBD-1)isconsideredaprominentAMPinepithelialdefenseagainstinfection.Highexpressionlevelshavebeenfoundinthekidneyandfemalereproductivetract,especiallyinpregnantwomen.Besidesantimicrobialactivity,hBD1hasaneffectonchemoattractiveimmaturedendriticcellsandprobablymemoryTcells.Afterreductionofdisulphide-bridgeshBD-1becomesapotentantimicrobialpeptideagainsttheopportuNISTicpathogenicfungusCandidaalbicansandagainstanaerobicGrampositivecommensalsofBifidobacteriumandLactobacillusspecies.
