ThemonoclonalantibodyHM102recognizestheextracellularpartofmembrane-boundTNF-RIIaswellasthesolubleformofTNF-RIIwhichisgeneratedbyproteolyticcleavageoftheextracellulardomain.ThesolubleformcanstillbindTNF-alphawithhighaffinityandfunctionsasaTNF-alphaantagoNIST.TNF-alphaisanimportantsignallingproteinintheimmunesystemwhichcanactivateinflammatoryresponses,induceapoptosis,regulatecellularproliferation,andmayevenpromotecancerprogression.TNF-alphacanbindtotwostructurallydistinctmembranereceptors,TNF-RIandTNFRII,whichhavebothdistinctandoverlappingdownstreamsignalingcascades.TNFRIisbelievedtobeexpressedonnearlyallcelltypes,whereasTNFRIIexhibitsmorerestrictedexpression,beingfoundoncertainsubpopulationsofimmunecellsandseveralothercelltypes.AdominantroleofTNFRIIhasbeenshowninthymocyteactivationbyTNF-alpha,whereasinductionofcytotoxicityandotherfunctionsaremediatedlargelybyTNF-RI.TNF-RIisequallywellactivatedbyboththe17kDasolubleand26kDamembrane-boundform,whereasTNF-RIIisactivatedonlybythemembraneboundformofTNF-alpha.Theantibodyisaagonisticreceptormodulatingantibody.ItenhancesinvitroTNFalpharesponsesbyincreasingtheaffinityofthesolubleformofTNF-alphaforTNF-RII.
