Themonoclonalantibody265-3K1recognizeshumanleukocyteelastase.Leukocyteelastase,amajorserineproteinaseinman,ispredominantlypresentintheazurophilicgranulesofneutrophilsandmonocytes.Elastasehasabroadrangeofextracellularmatrixsubstratesincludingelastin,proteoglycans,collagenandfibronectin.Theactionofelastaseiscontrolledbyserineproteinaseinhibitors.Elastase,whenreleasedduringinflammation,israpidlyboundbyitstwomaininhibitors,alpha1-PIandalpha2-macroglobulinetoformelastase-inhibitorcomplexes.Inadditionmucosasecretionsmaycontainthelocallysecretedelastaseinhibitorselafin/SKALPandSLPI.Whensecretedatsitesofinflammationelastasecancauseseveretissuedamage.Animportantrolehasbeensuggestedforhumanelastaseinvariousinflammatorydisorders,includingpulmonaryemphysema,sepsis,arthritis,nephritisandcertainskindiseases.ElastaseinducestheproductionofIL-8inhumanbronchialepithelial,aprocesthatoccursinpartthroughTLR4.
