Themonoclonalantibody2D4reactswithratmyeloperoxidase(MPO).MPOisaglycoproteinwithanalpha2beta2heteromultimerexpressedinallcellsofthemyeloidlinage.MPOisabundantlypresentinazurophilicgranulesofpolymorphonuclearneutrophils.Itisanimportantenzymeusedduringphagocyticlysisofengulfedforeignparticleswhichtakespartinthedefenseoftheorganismthroughproductionofhypochlorousacid(HOCl),apotentoxidant.MPOisrapidlyreleasedbyactivatedpolymorphonuclearneutrophils.InvolvementofMPOhasbeendescribedinnumerousdiseasessuchasatherosclerosis,lungcancer,Alzheimer"sdiseaseandmultiplesclerosis.AutoimmuneantibodiestoMPOareinvolvedinWegenersdisease.SincethediscoveryofMPOdeficiency,initiallyregardedasrareandrestrictedtopatientssufferingfromsevereinfections,MPOhasattractedmoreclinicalattention.InexperimentalstudiesantibodiestoMPOcanbeusedforvariouspurposesrangingfromflowcytometricanalysistodetectionofpolymorphonuclearneutrophilsintissuesections.
