ThemonoclonalantibodyVIF9recognizestheratsurfactantproteinD(SP-D).SP-Dbelongstothecollectinfamiliy.Theseproteinsareoligomericproteinscomposedofcarbohydrate-recognitiondomains(CRD)attachedtocollagenousregions.TheyarestructurallysimilartotheficolinsalthoughtheymakeuseofdifferentCRDstructures:C-typelectindomainforthecollectins.Theanti-microbialeffectormechanismsofSP-Daredirectopsonization,neutralization,andagglutination.Thuslimitingtheinfectionandconcurrentlyorchestratingthesubsequentadaptiveimmuneresponse.ThelungisthemajorsiteofsynthesisofSP-D,wherethemoleculesareproducedandsecretedontotheepithelialsurfacebyalveolartypeIIcellsandunciliatedbronchialepithelialcells.SP-Disalsofoundindifferentepithelialcellsofthegastrointeststinaltractandinepithelialcellsofexocrineglands.SP-Dsynthesisandsecretionincreasesignificantlyafterinflammatorystress.IncreasedamountsofSP-Dinlavageandtissue,particularlyintypeIIpneumocytes,inClaracellsandinhyperplasticgobletcellsarefoundininflamedlungs.ThelocalizationofSP-Dinendocyticvesiclesandinlysosomalgranulesofalveolarmacrophagessuggeststhatareceptor-mediateduptakeoccurs.SP-Dbindstoapoptoticneutrophilsandenhancestheirclearancebyalveolarmacrophages.MonoclonalantibodyVIF9specificforratsurfactantproteinDshowssignificantcrossreactivitywithhumanSP-D.MonoclonalantibodyVIF9stainsparaffinsectionsofratlung.Itcannotbeusedforstainingofparaffinsectionsofhumanlung.ItcanalsobeusedasdetectionantibodyforELISAonwellscoatedwithhumanSP-D.
