ThemonoclonalantibodyWU13-15recognizesaneoepitopeonthe61kDacomplementcomponentC9,integratedintheterminalcomplementcomplex(TCC).Thecomplementsystemisanancientproinflammatoryandmicrobialdestructionsystem,thatmaybeconsideredpartofboththeinnateandadaptiveimmunesystems.Itconsistsoftheclassical,alternative,andlectin-bindingpathways.Eachpathwayistriggeredinadistinctmanner,yetalldepositC3fragmentsonatargetandengageacommonterminalsequencecalledTCCorthe"membraneattackcomplex"(MAC).Incontrasttotheactivationpathways,whichrequireenzymaticcleavageforactivation,theterminalpathwayreliesonconformationalchangesinducedbybindingofthedifferentsubunits.TCCiscomposedofacomplexoffourcomplementproteins(C5b,C6,C7,andC8)whichbindtotheoutersurfaceofthetargetplasmamembrane,andmanycopiesofafifthprotein(C9)thathookuptooneanother,formingaringinthemembrane.TheringstructureformedbyC9isaporeinthemembranethatallowsfreediffusionofmoleculesinandoutofthecell.Ifenoughporesform,thecellisnolongerabletosurvive.Themembraneattackcomplexisinitiatedwhenthecomplementprotein,C5convertase,cleavesC5intoC5aandC5b.BindingofC6facilitatesbindingofC7whichalterstheconformationofthecomplex.AfterbindingofC8,avariablenumberofC9moleculesassociatewiththeC5b678complex,togetherconstitutingTCC.TheformationofTCCcauseslysisofcellsorcantriggeravarietyofcellularmetabolicpathwaysresultinginthesynthesisandreleaseofinflammatorymediators.TCCcontainsneoantigensthatareabsentintheindividualnativeC9proteins.NeoantigensarepresentbothinthemembraNEBound(MAC)andthefluidphase(SC5b-9)complex.TCCispresentinnormalhumanplasmaandincreasesuponcomplementactivation.
