ThemonoclonalantibodyMNB4(previouslyknownasclone20)recognizeshumanpentraxin3(PTX3),belongingtothelongpentraxinfamily.PTX3isanacute-phaseglycoproteinof~45kDawithglycosylationaccountingforabout10%ofitsmolecularweight.PTX3hasacomplexoligomericstructurewithprotomerslinkedtoeachotherbydisulfidebonds.PTX3expressionistriggeredbyinflammatorycytokines,resultinginhigherlevelsofcirculatingPTX3.SeveralcelltypeshavebeenreportedtoproducePTX3,namelymacrophages,endothelialcells,neutrophilsandsynoviocytes.PTX3isinvolvedinhostdefenseagainstpathogeninfection,intheregulationofthescavengeractivityofmacrophagesanddendriticcells,andinmodulationofcomplementactivitybybindingtoC1q.FurThermore,PTX3hasbeenimplicatedinmatrixdepositionofcumuluscells.Moreover,PTX3interactswithotherBIOLOGicallyactivemolecules,causingtheirfunctionalblockade.Thishasbeendemonstratedforfibroblastgrowthfactor-2(FGF-2),forwhichPTX3actsasaninhibitor,leADIngtoinhibitionofangiogenesis.PTX3,likeotherpentraxinsC-reactiveprotein(CRP)andserumamyloidPcomponent(SAP),bindsapoptoticcellsanddebris.PTX3isusefulasanearlyindicatorofmyocyteirreversIBLeinjuryinischemiccardiomyopathy.PTX3isnotonlyinvolvedininflammatoryvesseldiseasesrelatedtoatherosclerosis,butalsoinpre-eclampsiaandsystemicsmallvesselANCA-associatedvasulitis,inwhichneutrophilsarekeyplayers.Therelationshipbetweentissuedamageandpentraxingenerationisstringentinacuteinjuries.PTX3tunesself-non-selfdiscriminationandtissuerepairduetotherecognitionofdiverseligandsbyPTX3andthroughregulationofeffectorpathways.
