Themonoclonalantibody8C8recognizeshumanalpha-defensin5(HD5).HD5isanantimicrobialpeptideproducedbyPanethcells,specializedepithelialcellsofthesmallintestinalcrypt.HD5isstoredinthePanethcellsaspropeptide(aa20-94)andreleasedasmaturepeptideaftertrypsindigestion,whichisco-secretedfromthePanethcellgranules.ThematureHD5(aa63-94)isprotectedfromfurtherdigestionbytheproteolysisenzyme.HD5functionsinmaintainingthecorrectmicrobialbalanceintheintestine.TransgenicmicethatexpressHD5areprotectedfromlethalchallengecausedbySalmonellatyphimurium.ExpressionlevelsofHD5arenegativelycorrelatedtointestinalinfection.HD5isausefulMarkerforintestinalmetaplasiaoftheuppergastrointestinaltract.HD5expressionhasalsobeendetectedintheupperhalfofthestratifiedsquamousepitheliumofthevaginaandectocervixwithincreasingintensitytowardthelumen.SecretedHD5isdetectableincervicovaginallavages,withhighestconcentrationsduringthesecretoryphaseofthemenstrualcycle.Themonoclonalantibody8C8wasraisedagainstrecombinantHD5(aa20-94)andrecognizesthepropeptideandthepartiallyprocessedforms(aa36-94and56-94),butithardlyrecognizesmaturepeptide.NocrossreactivitywithHD6,lysozyme,orsPLA2couldbedetected.
