Themonoclonalantibody6G12reactswithhumanMASP-2andhumanMAp19.MASP-2isatrypsin-likeserineproteaseandplaysanimportantroleintheinitiationoftheMBLcomplementactivationpathway.Threepathwaysofcomplementactivationhavebeenreported:theantibody-dependentclassicalpathway,theantibody-independentalternativepathwayandthelectinpathway.Activationofeachpathwayinvolvesformationofserineproteasecomplexes,whichresultsinactivationofthecentralcomplementcomponentC3.Inthelectinpathway,mannosebinding-lectin(MBL)-associatedserineproteases(MASPs)formcomplexeswithpolymericlectinmoleculeswhichareinvolvedinpatternrecognition.Uponbindingoftherecognitionmoleculestocarbohydratesonthesurfaceofmicroorganisms,MASPsareconvertedtotheiractiveformsandinitiatecomplementactivation.ThreetypesofhumanMASPhavebeenreported.MASP-1,MASP-2andMASP-3.Mannan-bindinglectin(MBL)andficolins,incomplexwithMBL-associatedserinproteases(MASPs),arecapableofactivationthecomplementsystem,thusmediatingthedestructionofinfectiousagents.MASP-2cleavesC4andC2andiscrucialfortheactivationofdownstreamcomplementcomponents.MAp19isanalternativesplicingproductoftheMASP-2gene.MAp19comprisesthefirsttwodomainsofMASP-2followedbyanextrasequenceoffouruniqueaminoacids(EQSL)atitsC-terminal.MASP-2andMAp19havebeenreportedtobindtoMBLinacalcium-dependentmanner.
