Themonoclonalantibody8B5reactswiththeCCP1/2-SPfragmentofhumanMASP-2.MASP-2isatrypsin-likeserineproteaseandplaysanimportantrolintheinitiationoftheMBLcomplementactivationpathway.Threepathwaysofcomplementactivationhavebeenreported:theantibody-dependentclassicalpathway,theantibody-independentalternativepathwayandthelectinpathway.Activationofeachpathwayinvolvesformationofserineproteasecomplexes,whichresultsinactivationofthecentralcomplementcomponentC3.Inthelectinpathway,mannosebinding-lectin(MBL)-associatedserineproteases(MASPs)formcomplexeswithpolymericlectinmoleculeswhichareinvolvedinpatternrecognition.Uponbindingoftherecognitionmoleculestocarbohydratesonthesurfaceofmicroorganisms,MASPsareconvertedtotheiractiveformsandinitiatecomplementactivation.ThreetypesofhumanMASPhavebeenreported.MASP-1,MASP-2andMASP-3.Mannan-bindinglectin(MBL)andficolins,incomplexwithMBL-associatedserinproteases(MASPs),arecapableofactivationthecomplementsystem,thusmediatingthedestructionofinfectiousagents.MASP-2cleavesC4andC2andiscrucialfortheactivationofdownstreamcomplementcomponents.
