ThemonoclonalantibodyMU14-2A5recognizesS100A9.Thecalcium-binding,migrationinhibitoryfactor-relatedproteins,MRP-8(S100A8)andMRP-14(S100A9)belongtotheS100proteinfamily.MRP14isco-expressedwithhumanMRP8,ahomologueproteininmyeloidcells,andplaysanessentialroleincalciumdependentfunctionsduringinflammation.ThisroleincludestheactivationofMac-1,theβ2integrinwhichisinvolvedinneutrophiladhesiontoendothelialcells.MRP14isdistinguishedfromotherS100memberproteinsbyitslongC-terminalregion.
TheexpressionofMRP-8(S100A8)andMRP-14(S100A9)islargelyconfinedtothecytosolofneutrophilsandmonocytes.Thecomplexformationoftheseproteinsisacalcium-dependentprocess.TheS100A8/A9heterocomplex,alsocalledMRP-8/MRP-14complexorcalprotectin,comprises60%ofthecytoplasmicproteinfractionofcirculatingpolymorphonucleargranulocytesandisalsofoundinmonocytes,macrophagesandilealtissueeosinophils.Ininflammatoryconditionssmallvenuelesstainwithbothanti-S100A8andS100A9.Thestainingofthetwosubunitsisalwayscoincident.TheS100A8/A9complexhasantibacterial,antifungalandimmunomodulatingandantiproliferativeeffects.Besidesthisitisapotentchemotacticfactorforneutrophils.
