Themonoclonalantibody80M2recognizestheextracellularpartof-membrane-boundTNF-RIIaswellas-thesolubleformofTNF-RIIwhichisgeneratedbyproteolyticcleavageoftheextracellulardomain.ThesolubleformcanstillbindTNF-alphawithhighaffinityandfunctionsasaTNF-alphaantagoNIST.-TNF-alphaisanimportantsignalingproteinintheimmunesystemwhichcanactivateinflammatoryresponses,induceapoptosis,regulatecellularproliferation,andmayevenpromotecancerprogression.TNF-alphacanbindtotwostructurallydistinctmembranereceptors,TNF-RIandTNF-RII,whichhavebothdistinctandoverlappingdownstreamsignalingcascades.TNFRIisbelievedtobeexpressedonnearlyallcelltypes,whereasTNFRIIexhibitsmorerestrictedexpression,beingfoundoncertainsubpopulationsofimmunecellsandseveralothercelltypes.AdominantroleofTNF-RIIhasbeenshowninthymocyteactivationbyTNF-alpha,whereasinductionofcytotoxicityandotherfunctionsaremediatedlargelybyTNF-RI.TNF-RIisequallywellactivatedbyboththe17kDasolubleand26kDamembrane-boundform,whereasTNF-RIIisactivatedonlybythemembraneboundformofTNF-alpha.Theantibodyisanon-agonisticreceptormodulatingantibody.ItenhancesinvitroTNFalpharesponsesbyincreasingtheaffinityofthesolubleformofTNF-alphaforTNF-RII.
