ThemonoclonalantibodyJL-1recognizesthecollagen-likeregion(CLR)ofmouseC1q,a459kDamoleculeconsistingofthreeindividualpolypeptidechains.TheantibodyhasbeengeneratedbyimmunizationofC1q-/-C57BL/6micewithpurifiedmouseC1q.
C1qformstogetherwithC1randC1stheC1macromolecule,thefirstcomponentoftheclassicalcomplementpathway.InteractionofimmunecomplexeswithC1qinducesaconformationalchangewithintheC1complex,whichresultsinactivationoftheclassicalpathway.C1qfunctionsasrecognitionunitbybindingtotheheavychainofIgGorIgM(FcgammaandFcmicro)providedthattheimmunoglobulinsareboundtotheirantigen.FurThermore,C1qcanalsorecognizemolecularpatternsassociatedwithpathogensanditcanbindtoapoptoticblebs,whereitactivatestheclassicalcomplementpathwayandmediatesphagocytosis.Assuch,C1qpromotestheclearanceofapoptoticcellsandsubsequentexposureofautoantigens,therebypreventingstimulationoftheimmunesystem.
C1qispredominantlyproducedbymacrophagesbutalsobyfolliculardendriticcells,interdigitatingcellsandcellsofthemonocyte-macrophagelineage.C1qdeficiencyhasaprofoundeffectonhostdefenceandclearanceofimmunecomplexes.AbsenceofC1qmaycauseautoimmunitybyimpairmentoftheclearanceofapoptoticcells.InheritedC1qdeficiencyisalsoassociatedwiththedevelopmentofsystemiclupuserythematosus(SLE).
ThemonoclonalantibodyJL-1isreactivewiththecollagen-likeregion(CLR)only,whichisthesameregiontowhichautoantibodiesinmiceandhumansarebinding.Anti-C1qautoantibodiesdepositinglomerulitogetherwithC1qbutinduceovertrenaldiseaseonlyinthecontextofglomerularimmunecomplexdisease.Thisprovidesanexplanationwhyanti-C1qantibodiesareespeciallypathogenicinpatientswithSLE.
