Themonoclonalantibody23C11recognizesoxidizedlow-densitylipoproteinreceptor-1(LOX-1).LOX-1isasingle-passtypeIImembraneprotein(~45kDa)andbelongstotheC-typelectin-likeproteinsuperfamily.LOX-1isexpressedathighlevelinendothelialcellsandvascular-richorganssuchasplacenta,lung,liver,brainaorticintima,bonemarrow,spinalcordandsubstantianigra.Itisalsoexpressedonthesurfaceofdendriticcells.ThisuniquescavengerreceptorLOX-1playsimportantrolesinatherogenesis.LOX-1mediatestherecognition,internalizationanddegradationofoxidativelymodifiedlowdensitylipoprotein(oxLDL)byvascularendothelialcells.OxLDLisaMarkerofatherosclerosis,inducingvascularendothelialcellactivationanddysfunction,resultinginpro-inflammatoryresponses,pro-oxidativeconditionsandapoptosis.LOX-1associateswithoxLDLinducingtheactivationofNF-kappa-Bthroughanincreasedproductionofintracellularreactiveoxygenandavarietyofpro-atherogeniccellularresponsesincludingareductionofnitricoxide(NO)release,monocyteadhesionandapoptosis.InadditiontobindingoxLDL,LOX-1actsasareceptorfortheHSP70proteininvolvedinantigencross-presentationtonaiveT-cellsindendriticcells,therebyparticipatingincell-mediatedantigencross-presentation.LOX-1isinvolvedintheinflammatoryprocess,byactingasaleukocyte-adhesionmoleculeatthevascularinterfaceinendotoxin-inducedinflammation.LOX-1alsoactsasareceptorforadvancedglycationend(AGE)products,activatedplatelets,monocytes,apoptoticcellsandbothGram-negativeandGram-positivebacteria.TheLOX-1geneisaso-calledimmediateearlygenethatisdynamicallymodulatedbyseveralfactorsinvitroandinvivo.LOX-1expressionisinducedbystimulisuchasinflammatorycytokines,OxLDL,TNF-alpha,TGF-beta,andANGIIinvitro,andseveralproatherogenicfactorsinvivo.Monoclonalantibody23C11neutralizesLOX-1andinhibitsHsp70bindingtodendriticcellsandHsp70-inducedantigencross-presentation.Invivo,targetingLOX-1withatumorantigenusinganti-LOX-1antibody23C11inducesanti-tumorimmunity.
