ThemonoclonalantibodyM.Ab.F11recognizesjunctionaladhesionmolecule-A(JAM-A)alsoknownasthehumanplateletF11-Receptor(F11R)andJAM-1isacelladhesionmolecule(CAM).JAM-Aisamemberoftheimmunoglobulinsuperfamilyfoundonthesurfaceofhumanplateletsandatintercellularjunctionsofendothelialcellsandepithelialcells.JAM-AbelongstogetherwithJAM-C(JAM-2)andJAM-B(VE-JAMorJAM-3)toafamilyofadhesionproteinswithaV-C2immunoglobulindomainorganization.JAMplaysanimportantroleintightjunctionswhereitisinvolvedincell-to-celladhesionthroughhomophilicinteraction.ItcodistributeswithothertightjunctioncomponentsasZO-1,7H6antigen,cingulinandoccludin.JAM-Aplaysaroleinplateletaggregation,secretion,adhesion,spreADIng.IntheplateletF11R/JAM-Aisamembraneproteininvolvedin2distinctprocessesinitiatedontheplateletsurface.Antibody-inducedplateletaggregationandsecretionbothdependentonFcgammaRIIandGPIIb/IIIaintegrin,aprocessthatmaybeinvolvedinpathophysiologicalprocessesassociatedwithcertainthrombocytopenias.AntibodymediatedplateletadhesionindependentfromFcgammaRIIorfibrinogenreceptorandthatappearstoplayaroleinphysiologicalprocessesassociatedwithplateletadhesionandaggregation.AphysiologicalrolefortheF11R/JAM-Aproteinwasdemonstratedbyitsphosphorylationafterthestimulationofplateletsbythrombinandcollagen.ApathophysiologicalrolefortheF11R/JAM-AwasrevealedbydemonstratingthepresenceofF11R/JAM-Aantibodiesinpatientswiththrombocytopenia.AdhesionofplateletsthroughtheF11Rresultedineventscharacteristicoftheactionofcelladhesionmolecules(CAMs).RecentdatasuggestsaroleforF11R/JAM-Aintheadhesionofplateletstocytokine-inflamedendothelialcellsandthusinthrombosisandatherosclerosisinducedinnon-denudedbloodvesselsbyinflammatoryprocesses.
