ThemonoclonalantibodyF18recognizesandneutralizesbothnaturalandrecombinantmousealphaInterferon(IFN-α).IFN-αisacytokinethatbelongstothetypeIinterferons(IFN-I).IFN-αissecretedbymanycelltypesincludinglymphocytes(NKcells,B-cellsandT-cells),macrophages,fibroblasts,endothelialcells,osteoblasts,microgliaandothers.InterferonsstimulatebothmacrophagesandNKcellstoelicitananti-viralresponse,andarealsoactiveagainsttumors.AlthoughallcellscanproduceIFN-I,plasmacytoiddendriticcells(pDCs)produce1,000-foldhigherlevelsthanothercelltypes,andareresponsIBLeforsystemicIFN-Iresponsestomanyviruses.TheyarecoinedasthenaturalIFN-producingcells.However,underdeprivedpDCcondition,otherdendriticcellsarecapableofproducinghighlevelsofIFN-I.
InterferonswereinitiallycharacterizedfortheirABIlityto‘interfere"withviralreplication,slowcellproliferation,andprofoundlyalterimmunity.IFN-αhasseveralregulatoryrolesanddiverseBIOLOGicalactivities,includingcontrolofcellularandhumoralimmuneresponses,inflammation,andtumorregression.Inaddition,IFN-αparticipatesintheregulationofvariouscellularandhumoralprocessessuchastheendocrinesystemmodulatesbehavior,brainactivity,temperature,glucosesensitiveneurons,feedingpatternandopiateactivity.WiththeavailabilityofmonoclonalantibodiesdirectedagainstIFN-α,itispossibletointerpretresultsobtainedfromcrudematerialscontainingbothIFN-αandIFN-β.Thedifficultiesinstudyinginvitroandinvivoeffectsof‘type1’InterferonsarisefromthefactthatbothalphaandbetaInterferonsareproducedinresponsetothesamestimuliandalsoseemtoactviathesamereceptor.TheseInterferonactivitiescanonlybedistinguishedfromoneanotherbyuseofspecificneutralizingantibodies.
